摘 要

目的：本论文建立了注射用磷丙泊酚钠血管刺激性试验的方法，对注射用磷丙泊酚钠进行局部毒性试验。方法：注射用磷丙泊酚钠血管刺激性试验采用5只成年健康新西兰兔，其中三只♀、两只♂。通过兔子左边耳朵的耳缘静脉推注供试品，其推注的剂量是每公斤35mg，给药浓度是每毫升35mg，给药容积为每公斤1ml，右边耳朵耳缘静脉推注等体积灭菌注射用水。一天给予药一次，连续注射3天，最后一次给药之后72小时解剖3只（2♀/1♂），剩余动物观查7天后解剖，肉眼和病理检察血管是否有部分变性或者坏死或者其他刺激性现象。结果：通过肉眼观察比较给药品组和对照品组，而对于血管处于给药的时期和恢复的时期都未看见溃烂、肿胀、红色斑点等一系列的刺激性症状；病理检查结果显示，在给药期及恢复期，每只兔耳3个取材点均未见血管内皮细胞肿胀、坏死、脱落，血管腔内无血栓，血管周围部分无炎症，无流血等病变。结论：该试验方法简单方便、准确、可靠，可以适合用作注射用磷丙泊酚钠血管刺激性试验。

Test for vascular irritation of fospropofol disodium for injection

Abstract

Objective: This paper established the irritation test with fospropofol disodium by intravascular injection, local toxicity test of fospropofol disodium for injection. Methods: In the vascular irritation test,we used 5 adult healthy New Zealand rabbits (3 female /2 male).Rabbits were injected by left ear margial vein with the dosage of 35mg/kg and the drug concentration of 35mg/ml,delivery volume was 1ml / kg, right ear was injected by margial vein with the same volume Sterile water for injection as the left ear.Once a day,continuous administration for 3 days, 72h after the last administration,3 rabiits (2 female / male) were dissected.The left animals were observed for 7 days,then dissected them and checked whether there is tissue degeneration or necrosis of irritant reactions by eyes and pathological examination.Results：Naked eye to observe the treatment group and the control groups of blood vessels in rabbit ear, in the dosing period and recovery period were no erythema, oedema, ulcer and irritative symptoms; pathological examination results show that during the dosing period and recovery period, each rabbit ear three sampling points showed no vascular endothelial cell swelling, necrosis and exfoliation, endovascular with no thrombus, perivascular tissue with no inflammation, no bleeding etc. lesions.Conclusion: the method is simple, accurate and reliable for vascular irritation of fospropofol disodium for injection.
