气相色谱法检测艾氟康唑原料药中有机溶剂残留的研究毕业论文

摘 要
目的:建立检测康唑中乙醇、异丙醇、乙腈、甲基叔丁基醚、乙酸乙酯、四氢呋喃、苯、三乙胺、正庚烷、甲苯、乙酸11种溶剂残留的气相色谱分析法。
方法:检测器为氢火焰离子化检测器,载气为N2,采用Agilent DB-624色谱柱(30m×0.32mm×1.8μm),以N,N-二甲基甲酰胺为溶剂,采取静态顶空气相色谱法,柱温为程序升温(初始40℃,维持10.0min,以20℃/min升到180℃,维持3.0min)检测了艾氟康唑中11种残留溶剂的含量。
检测结论是乙醇、异丙醇、乙腈、甲基叔丁基醚、乙酸乙酯、四氢呋喃、苯、三乙胺、正庚烷、甲苯、乙酸在考察范围内线性关系良好,r2均大于0.99,平均回收率为99.4% ~ 101.9%,RSD在0.95%~ 3.44%的范围之内。检测限分别为330,400,340,110,400,300,30,3240,70,70,9000 ng/ml。
结论是本检测法操作方便、灵敏、准确、重现性好,可被很好的用在实际生产中的艾氟康唑残留溶剂的检查中。

Determination of residual solvents in efinaconazole by gas chromatography
Abstract
Objective :To establish a method for the determination of seven residual solvents in ramelteon.Those residual solvents are ethanol, isopropanol, acetonitrile, methyl tert butyl ether, tetrahydrofuran, ethyl acetate, benzene, three ethylamine, n-heptane, toluene and acetic acid.
Methods :Nitrogen gas was used as carrier gas. FID was used as detector.The residual solvents were separated on Agilent DB-624(30m×0.32mm×1.8μm)capillary chromatographic columnand the solvent was dimethylformamide.
Ethanol, isopropanol, acetonitrile, methyl tert butyl ether, tetrahydrofuran, ethyl acetate, benzene, three ethylamine, n-heptane, toluene and acetic acidin efinaconazoleusing temperature programming (Initial temperature was 40 ℃ and keep 10 minites ,increase the temperature from 40℃ to 180℃ at the rate of 20℃/min,thenkeep 3.0 minute)by static headspace gas chromatography.
Results :The good linearrelation ofethanol, isopropanol, acetonitrile, methyl tert butyl ether, tetrahydrofuran, ethyl acetate, benzene, three ethylamine, n-heptane, toluene and acetic acidether were achieved within the investigatedconcentration range and r2 were0.99.The average recoveries were rangedfrom 99.4% to 101.9%(The RSD were0.95%~3.44%).The limit of detection were separately 0.33,0.40,0.34,0.11,0.40,0.30,0.03,3.24,0.07,0.07 ng•ml-1and 9.00ng.
Conclusions:The method is simple, sensitive,accurate and reproducible. It can be used for detection of residual solvents in efinaconazole in practical production.

第一章 绪论
1.1 艾氟康唑药品概述
艾氟康唑(efinaconazole)是首个外用的三唑类抗真菌药物,商品名为Jublia,是由加拿大DOW Pharmaceutical制药公司研发,2014年6月6日经美国FDA批准上市。临床上用该药为质量分数10%的溶液来治疗主要用于局部涂抹治疗由红色毛癣菌、毛癣菌引起的手足癣(即灰指甲)。艾氟康唑的中文化学名称是(2R,3R)-2-(2,4-二氟苯基)-3-(4-亚甲基哌啶-1-基)-1-(1H-1,2,4-三唑-1-基)-2-丁醇[1][2],结构见图1-1。
