摘 要

盐酸奥洛他定是一个新型选择性二苯并氧杂环庚三烯 - 2 - 乙酸类组胺 H 1 受体抑制剂, 能够抑制速激肽及其它化学递质, 如组胺、花生四烯酸、血栓素、白三烯等的释放, 主要用于过敏性鼻炎、荨麻疹、伴有瘙痒的皮肤病(湿疹、皮炎、痒疹、皮肤瘙痒症、寻常银屑病、多形渗出性红斑)的治疗。目前，奥洛他定已具有多种剂型，常见用于治疗过敏性鼻炎的鼻腔喷雾剂以及治疗过敏性结膜炎的体征和症状的滴眼液。苯扎氯铵是 3 种季铵盐类同系物的混合物，常用作防腐剂。本课题要求熟练掌握压液相色谱仪等仪器的实验操作方法，并学会建立药物的质量研究方法，从而通过测定这 3 种同系物的量，确定奥洛他定鼻喷雾剂中苯扎氯铵的含量。

Olopatadine nasal spray in the determination of benzalkonium chloride content

Abstract

Olopatadine hydrochloride is a novel selective dibenzo oxetanyl cycloheptatriene - 2 - acetic acid class of histamine H 1 receptor antagonist, can inhibit tachykinin and other chemical mediators, such as histamine, arachidonic release acid, thromboxane, leukotrienes, etc., mainly for allergic rhinitis, urticaria, pruritus associated with skin diseases (eczema, dermatitis, prurigo, itchy skin disease, psoriasis vulgaris, multiforme exudative erythema) of treatment.Currently, olopatadine already has a variety of formulations, commonly used in the treatment of allergic rhinitis, nasal sprays, and treatment of allergic conjunctivitis signs and symptoms of eye drops.Benzalkonium chloride is a mixture of three kinds of quaternary similar homologue, commonly used as a preservative.This topic proficiency pressure liquid chromatography, and other instruments of the experimental methods of operation, and learn methods to establish quality of drugs, so by measuring the amount of these three kinds of homologues determine olopatadine nasal sprays benzalkonium chloride content.

第一章 前言

奥洛他定是由日本东京协和发酵工业株式会社研发并上市，具有广泛的药物作用，如组织胺H1受体拮抗剂作用，化学介质抑制作用，速激肽释放抑制作用和嗜酸粒细胞浸润抑制作用^[1]。且奥洛他定并无H₁受体拮抗剂类抗过敏药物常见的中枢神经系统抑制及心脏毒副作用^[2]。目前常见的奥洛他定有三种剂型，片剂、1%滴眼液和鼻腔喷雾剂。阿洛刻（Allelock）常被用来治疗变应性鼻炎、荨麻疹、皮肤病（湿疹、皮肤炎、痒疹、皮肤瘙痒症、寻常型干癣、多形性渗出红斑）伴发的瘙痒^[3]；而帕坦洛（Patanol）—1%滴眼液由日本爱尔康(Alcon)公司引入并开发，在治疗过敏性结膜炎的体征和症状的应用上具有显著效果^[4]。奥洛他定分别于 1996 年在美国、1997 年在加拿大曾做为过敏性结膜炎用药上市, 商品名为 Patanol 滴眼液, 2000 年 12 月又做为抗过敏药在日本上市, 国内正在研制开发阶段 。
