摘 要

富马酸喹硫平是最先由英国阿斯利康公司研制出来的一种抗精神病药，它作用于多种受体和神经递质，不仅可以减轻精神分裂症患者的多种症状，而且对于治疗他们的认知功能也有一定的疗效。本论文的目的在于研究富马酸喹硫平的处方和制备工艺，以及它的放大研究和稳定性考察，最后确定合理的处方。

目的：根据不同处方所制备的富马酸喹硫平片的质量及稳定性，对本品的处方和制备方法进行筛选优化，因此获得高质量的富马酸喹硫平片。

方法：基于富马酸喹硫平的物理化学性质和国内材料的供应情况，参照国内外上市品的处方组成，制定了富马酸喹硫平片的填充剂、粘合剂、崩解剂、润滑剂的种类和用量，通过常规的湿法制粒方法制得富马酸喹硫平片，为处方筛选设计不同的方案，通过一定的方法研究溶出度。

结果：富马酸喹硫平200 mg，乳糖16 mg，微晶纤维素16 mg，磷酸氢钙72 mg，10 %聚维酮K30 87 mg，羧甲基淀粉钠16 mg，硬脂酸镁12 mg，这样才与原研品的溶出曲线最为相似。

结论：富马酸喹硫平片的处方符合设计要求，制备工艺可行，制剂稳定，有着安全、效果显著的优点。

Study on the Formulation Technology of Quetiapine Fumarate Tablets

Abstract

The purpose of this paper is to study the preparation and preparation of quetiapine fumarate, as well as its amplification study and stability study, and finally to determine a reasonable prescription.

Aim: According to the quality and stability of quetiapine fumarate tablets prepared by different prescriptions, the prescription and preparation method of this product were screened and optimized, therefore, high quality quetiapine fumarate tablets were obtained.

Method: Based on the physicochemical properties of quetiapine fumarate and the supply of domestic materials, the filler, binder, disintegrant and lubricant of quetiapine fumarate tablets were prepared according to the prescription composition of domestic and overseas listed products. And the amount of quinapram fumarate was prepared by conventional wet granulation method. The dissolution rate was studied by a certain method.

Result: Paclitaxel fumarate 200 mg, lactose 16 mg, microcrystalline cellulose 16 mg, calcium hydrogen phosphate 72 mg, 10% povidone K30 87 mg, sodium carboxymethyl starch 16 mg, magnesium stearate 12 mg, This is only with the original product of the dissolution curve is most similar.

Conclusion:The formulation of quetiapine fumarate meets the design requirements, the preparation process is feasible, the preparation is stable, has the advantages of safety and remarkable effect.

1.1引言

精神分裂症是指从精神健康到可诊断的精神障碍的连续统一分布的亚临床精神病经验（可能包括奇怪的行为，奇怪的言语，不寻常的知觉经验和社会的缓解）。精神分裂症^[1]患者大多数都会表现出不同程度的思维混乱、情绪低下、精神错乱以及神志不清，在青壮年中发病率较高。现代社会的紧凑感越来越强，人们面临的各种压力也与日俱增，患有神经方面疾病的人越来越多，因此临床上关于抗神经疾病药物的需求仍然很大。目前，我国科研人员在临床上针对精神疾病的发病原因做了很多的探究，研究发现精神病患者发病大多数是由于中枢神经系统的5-羟色胺过于兴奋或者多巴胺^[2]过于低迷，这两者是精神病患者发病的主要原因。
