摘 要

 更年期综合症（简称MPS）是妇女常见疾病，指绝经前后出现性激素波动或减少所致的一系列以自主神经系统功能紊乱为主，伴有神经心理症状的一组症候群^[1]。益母草碱是益母草中主要的生物碱成分，具有调经止血，较强的子宫兴奋功能。因此，我们提出了一种假设，益母草碱对更年期的卵巢衰弱有改善作用。随后，我们设计了本实验，目的是为了观察益母草碱对卵巢功能衰退小鼠的改善效果，进行盐酸益母草碱的药效学验证。

方法：（1）分组分别注射环磷酰胺、二氧化乙烯基环己烯(VCD)建立小鼠卵巢功能衰退模型，使用酶标法检测小鼠血清的雌二醇水平，筛选出最合适的造模组。（2）通过连续15天腹腔注射环磷酰胺建立小鼠卵巢早衰模型，后期给予灌胃益母草碱进行治疗。（3）检测小鼠的雌二醇水平以及药物对卵巢，子宫，肾脏，肝脏器官组织的影响。

结果：经过15天的灌胃给予益母草碱，卵巢早衰小鼠血清中的雌二醇水平均有有明显的上升，FSH下降，且对脏器有良好的修复作用，其中30mg/kg剂量的益母草碱治疗效果最好。 结论：益母草碱对卵巢衰弱有所功效，益母草碱具有类雌激素样作用。

Pharmacodynamics of Leonurus Hydrochloride

Abstract

Menopausal syndrome (MPS) is a common disease in women. It refers to a series of disorders of autonomic nervous system with neuropsychological symptoms caused by fluctuation or decrease of sex hormones before and after menopause. During the onset of the disease, female patients'ability to regulate the body declines, which seriously endangers women's psychological and physiological health. Leonurine are the main alkaloids in Leonurus heterophyllus. Literatures and reports have pointed out that many studies on the pharmacological effects and mechanisms of Leonurineare different from traditional indications. It has the function of exciting uterus. Therefore, we propose a hypothesis that Leonurine can improve ovarian failure in menopause. Subsequently, we designed this experiment in order to observe the improvement effect of Leonurus alkaloids on ovarian dysfunction mice, and to verify the pharmacodynamics of Leonurus alkaloids hydrochloride.

Methods: (1) Cyclophosphamide and Vinyl Cyclohexene Dioxide (VCD) were injected into groups to establish ovarian dysfunction model in mice. Serum estradiol levels were detected and the most suitable model group was selected. (2) The mice were injected with cyclophosphamide for 15 days to establish the model of premature ovarian failure, and then administered with Leonurus alkaloids. (3) Detection of estradiol levels and their effects on ovaries, uterus, kidneys and liver tissues. Result: After 15 days of administration, the estradiol level of premature ovarian failure mice increased significantly, and had no significant effect on organ index. Among them, 30 mg/kg of Leonurine had the best therapeutic effect.

Conclusion: Leonurine have some effects on ovarian weakness, and Leonurine have estrogen-like effects.

1.1 更年期与卵巢早衰

围绝经期综合征（MPS）简称更年期，是妇女常见疾病。指妇女绝经前后出现的一系列因性激素波动或减少所导致的神经系统功能紊乱，并伴有心理症状的症候群^[1]，其主要的表现为女性的内分泌功能紊乱、代谢异常，引发如潮热、性欲低下、心烦易怒等一系列生理、心理症状。更年期症状多发于45-55岁的女性，由于生理性、病理性或手术等而引起的卵巢功能衰退。患者也有可能会因为情绪变化，出现提前发病的现象。更年期症状持续时间因个体差异不尽相同。女性进入更年期后，生理功能逐渐衰退，机体调节能力严重下降，更容易受外界环境因素刺激^[2]，影响正常的工作与生活。
