氧化魔芋壳聚糖-胶原蛋白肽水凝胶的制备和性能研究毕业论文

摘 要

壳聚糖是甲壳素在碱性条件下脱乙酰基的产物,因此来源十分广泛。此外,它因为具有良好生物相容性、生物降解性、无毒、易于改性等特性,所以广泛应用于医疗、食品、生物工程、纺织等领域。但由于壳聚糖不溶于中性和碱性溶液,使其应用受到了极大的限制。本论文中,我们将胶原蛋白肽接枝到壳聚糖上,以改善壳聚糖的性能。魔芋葡甘聚糖是水溶性、非离子型高分子天然多糖。由于它良好的生物相容性,生物降解性以及优异的凝胶特性,近些年来,KGM作为生物医用材料在控制释放系统中已经展现出良好的前景。本论文中,我们使用高碘酸钠对魔芋葡甘聚糖进行氧化并作为一种新型的交联剂,以代替壳聚糖材料中经常使用的戊二醛、甲醛等具有毒性的交联剂。本论文研究的目的是制备出具有不同比例的、无毒的、可生物降解的氧化魔芋葡甘聚糖-壳聚糖-胶原蛋白肽水凝胶,然后研究它们物理、化学、机械、形态特性,这些特性的评估能够提供有价值的信息,进而去了解水凝胶的形成机理和应用前景。因为氧化魔芋葡甘聚糖-壳聚糖-胶原蛋白肽水凝胶的制备过程简单、可行,并且可以在温和的条件下进行,而且不使用任何外来的有毒的交联剂,如乙二醛、戊二醛、和二环氧化合物等,我们相信这样一个复合材料会在创面处理、药物输送、组织应用工程等相关领域有着很好的应用前景。

Abstract

Chitosan is a deacetylation product of chitin in alkaline environment.Therefore,it has wide source.Besides,it can be widely used in medical care, food,bioengineering ,textile and other fields,because of its better biocompatibility, biodegradability, non-toxicity, and easily modified ability.But,chitosan is insoluble in neutral and alkaline solution,which limits its application greatly.In this paper,we grafted collagen peptides onto chitosan to improve the characteristics of Chitosan.KGM is high-molecular weight water-soluble non-ionic natural polysaccharide. Due to its good biocompatibility, biodegradability as well as excellent gel-forming properties, KGM already shows promise in controlled release systems serving as biomedical materials in recent years;In this paper ,KGM is oxidized with sodium periodate and becomes a novel cross-linking agents to replaces glutaraldehyde and formaldehyde which were most common and poisonous cross-linking agents used in various CS-based biomaterial formulations.The aim of the paper is to prepare a non-toxic,, biodegradable oxidized KGM- chitosan -collagen peptides hydrogels with different mixing ratios, and then to investigate their physical, chemical, mechanical, morphological characteristics. The evaluations of these properties contribute to some meaningful information to understand the formation mechanism of hydrogels and application prospect.Since this process of the oxidized KGM- chitosan -collagen peptide hydrogel formation is simple, feasible, and usually performed under mild conditions without employing any extraneous toxic cross-linking agents, such as glyoxal, glutaraldehyde, and diepoxy compound etc., we believe that such a composite matrix would have potential applications in wound management, drug delivery, tissue engineering, and other related fields.
